First, I'll ask them politely to leave. If they refuse, I'll then tell them to get out or I'll call the police. In my state, you're officially trespassing when someone asks you to leave and you don't.
I'm a very private, solitary person at home. If someone I don't feel comfortable staying with for a long period of time, I'll find ways to gently encourage them that it is time to go. If it's someone that I don't like, I'll come out and say it in plain terms that I would like them to leave.
I rarely have any guests over. I had a girlfriend ask me to leave very politely. She said in the most sweetest and flirtatious voice "I'm gonna have to kick you out now." and I politely obliged. It was a one night stand. I never saw her again.
